The JAK/STAT pathway is one of the important signaling pathways that regulate cell proliferation, differentiation, migration and apoptosis . At the same time, the JAK/STAT pathway can regulate the expression of the downstream anti-apoptotic protein Bcl-2 and cause the inhibition of apoptosis . In this study, we found that after xanthohumol acted on human squamous cell carcinoma A431 cells, the protein expressions of JAK2, STAT3 and Bcl-2 in the cells showed a decreased trend. Therefore, we speculated that the anti-xanthohumol effect on human squamous cell carcinoma A431 cells might be realized by inhibiting the expression of JAK2, STAT3 and Bcl-2 proteins.

In conclusion, xanthohumol can inhibit the growth, migration and invasion of A431 cells, and promote the apoptosis of A431 cells by inhibiting the expression of JAK2, STAT3 and Bcl-2 proteins, which provides a certain experimental basis for the clinical application of xanthohumol as a potential anti-squamous cell carcinoma drug.
